Is there any possible way that I can secure the NasLite server from other users on the same network?

the level of security you are talking about is not what Naslite was designed for.

If security is a driver for you then go for a main stream distro.

Any Linux distro can be stripped down to run with minimal resources - or you could go for one like Feather or Damn Small Linux. Most distros come with Samba (SMB) already installed, you will have to do some configuration to get passworded access running. It might also be worth keeping a watch on Slax since they are working on a live CD server.
